Contrave is a prescription weight-loss tablet that combines two medications: naltrexone and bupropion. Together, they work on the brain to reduce hunger and control cravings. It is typically prescribed alongside a reduced-calorie diet and increased physical activity.
Contrave is a fixed-dose combination of naltrexone hydrochloride (an opioid antagonist, 8 mg) and bupropion hydrochloride (a norepinephrine-dopamine reuptake inhibitor, 90 mg). Naltrexone blocks beta-endorphin-mediated inhibition of pro-opiomelanocortin (POMC) neurons, while bupropion stimulates POMC neuronal firing, synergistically enhancing anorexigenic signalling in the hypothalamus. Phase III trials (COR) showed 5.4% placebo-subtracted weight loss at 56 weeks.
