Tesofensine

Tesofensine is an investigational weight loss medication that works by increasing the levels of three brain chemicals — serotonin, dopamine, and noradrenaline — which together reduce appetite and may slightly boost metabolism. It is taken as a once-daily tablet.

Tesofensine is a triple monoamine reuptake inhibitor (serotonin-noradrenaline-dopamine) originally developed for Parkinson’s and Alzheimer’s disease but repurposed for obesity after weight loss was observed as a side effect. Phase II trials demonstrated dose-dependent weight loss of 4.5–10.6% over 24 weeks versus 2.0% for placebo, primarily through appetite suppression and increased satiety. Phase III trials are ongoing; it is not yet approved by major regulatory agencies.
