Pramlintide is a synthetic analogue of the neuroendocrine hormone amylin, co-secreted with insulin from pancreatic beta cells. It acts as an amylin receptor agonist, delaying gastric emptying, suppressing postprandial glucagon secretion, and promoting satiety via central mechanisms in the area postrema of the brainstem. It is approved as an adjunct to insulin therapy in type 1 and type 2 diabetes and has demonstrated modest but statistically significant weight loss in clinical trials.
